Half Fraser (Demi-Fraser) Broth is used with ferric ammonium citrate for the selective enrichment of Listeria species and is not intended for use in the diagnosis of disease or other conditions in humans. Primary enrichment broth for the isolation of Listeria spp. formulated according to ISO 11290. The selective components acriflavine and nalidixic acid are blended into the base powder and the ferric ammonium citrate is added to the tempered broth after sterilization. Half Fraser (Demi-Fraser) Broth, NCM0001 was developed to give improved results for the isolation of Listeria spp. with both traditional and ELISA methods. The use of LSM is based on the method of isolating lymphocytes using density gradient centrifugation (differential centrifugation), resulting in the formation of several distinct cell layers. Mononuclear cells (lymphocytes and monocytes) and platelets will be contained in the banded plasma-LSM interphase due to their density. Erythrocytes and granulocytes will migrate through the gradient to the bottom of the tube. Lymphocytes can be recovered by aspirating the plasma layer and removing the cells.

Sabouraud Dextrose Agar is used for the cultivation of fungi. Conforms to Harmonized USP/EP/JP performance requirements and is not intended for use in the diagnosis of disease or other conditions in humans. A medium recommended by the Harmonized Pharmacopeia for the isolation and identification of Candida albicans from non-sterile samples. Conforms to USP/EP/JP performance specification. The medium is commonly abbreviated to SDA. The medium is also used for the preparation and maintenance of fungal test strains as described by the Harmonized Pharmacopeia. The peptone digests and dextrose provide a nutritious base for luxuriant fungal growth and the acidic pH affords selectivity against bacteria. A color change from purple to yellow indicates growth of a bile tolerant, lactose fermenting organism such as Escherichia coli. According to the Harmonized USP/EP/JP, MacConkey Broth is used as a selective enrichment broth, with subculture performed onto MacConkey Agar. RVS is not intended for use in the diagnosis of disease or other conditions in humans.Rappaport et al. originally described the broth, which was later modified by Vassiliadis et al. and optimized by van Schothorst et al. Enzymatic digest of soya provides a source of nitrogen and amino acids and sodium chloride provides osmotic pressure. The potassium phosphate components act as a buffer. Magnesium chloride and malachite green act as selective agents against non-target microorganisms.
